摘要:
以金针菇子实体为研究体系,多糖得率为指标,通过Box-Behnken试验设计,结合响应面分析法,建立回归方程,得到酶解预处理-微波辅助提取金针菇多糖的最佳工艺条件:果胶酶质量分数2.5%,pH=4,微波功率650 W,酶解温度40 ℃,酶解时间45 min,液固比25 mL/g,微波辐射150 s。在此条件下多糖得率可达(11.35±0.10)%,且扫描电子显微镜(SEM)照片显示,该工艺条件对金针菇组织结构破坏明显。
Abstract:
Through Box-Behnken design and regression analysis,a response surface method was used to optimize the yield of polysaccharides extracted from fruiting body of Flammulina velutipes.The yield could reach(11.35±0.10)% and the optimal extraction conditions were as follows:pectinase dosage of 2.5%,pH=4,microwave power of 650 W,enzymolysis temperature of 40 ℃,enzymolysis time of 45 min,liquid solid ratio of 25 mL/g,and radiation time of 150 s.The process caused distinct damage on scan electronic microscope(SEM)images of Flammulina velutipes.
