摘要:
为了探讨盐酸柯诺拉赞对SD大鼠中枢神经系统和Beagle犬心血管及呼吸系统的用药安全性,对大鼠单次灌胃给予超纯水,盐酸柯诺拉赞20、60、200 mg/kg和盐酸氯丙嗪20 mg/kg,进行功能观察组合试验,观察一般体征、行为方面、神经方面、自主体征等指标。犬单次经十二指肠给予超纯水,盐酸柯诺拉赞1、3、10 mg/kg,观察麻醉犬的心率、血压及呼吸等指标。结果表明:除200 mg/kg剂量外,其余剂量的盐酸柯诺拉赞对动物的神经系统、心血管系统及呼吸系统均无影响。在上述剂量下的盐酸柯诺拉赞药物安全性较好,具有良好临床应用前景。
Abstract:
To evaluate the safety of carenoprazan hydrochloride on nervous system of SD rats and cardiovascular system, respiratory system of Beagle dogs, SD rats were orally administrated with ultra-pure water, 20, 60, 200 mg/kg doses of carenoprazan hydrochloride and 20 mg/kg dose of chlorpromazine hydrochloride, respectively. The functional observation battery was performed on tested rats to observe indicators about general physical signs, behavioral aspects, neurological aspects and autonomous signs. Ultra-pure water and 1,3,10 mg/kg doses of carenoprazan hydrochloride were given to the anesthetized dogs. The heart rate, mean arterial pressure and other related indicators of the anesthetized dogs were observed. Results suggested that 20 and 60 mg/kg doses of carenoprazan hydrochloride had no significant effect on the nervous system of SD rats, 1, 3 and 10 mg/kg doses of carenoprazan hydrochloride had no significant effect on cardiovascular system and respiratory system of Beagle dogs. Under the conditions, carenoprazan hydrochloride had a good security except for 200 mg/kg in rats.
